Investigation of the Effect of Daily Oral Vitamin C in Preventing the Postoperative Adhesion Formation: An Experimental Study

Objective: Investigation of the effect of daily oral vitamin C in preventing the adhesion on uterine horn model in rats.

Material and Method: Twenty one female Wistar Albino rats were divided into three groups as random. Group 1 (n =7): Only abdomen was opened and closed. Group 2 (n =7): A 2 cm linear incision was made to the right uterine horn and closed with 4/0 vicryl. Group 3 (n =7): After the administration of vitamin C at a dose of 50 mg/kg a day before the operation, a 2 cm linear incision was made to the right uterine horn and closed with 4/0 vicryl. All rats were opened after 15 days. Adhesion scoring and severity were evaluated according to Lisky scale. The right uterine horn was removed and fixed under 10% formaldehyde. Masson’s trichrome, TUNEL and Immunohistochemical staining were performed on 5 μm sections taken from paraffin blocks. Vascular endothelial growth factor (VEGF), angiogenesis, Type I collagen and malondialdehyde (MDA) were performed histologically in the traumatized uterine horn region.

Results: Adhesion area and adhesion severity were significantly lower in G3 than G2. When G2 and G1 were compared, fibrosis, angiogenesis, VEGF, Type 1 collagen and MDA scores were significantly higher in G2. VEGF and type 1 collagen score were significantly higher in G3 than G1. Fibrosis, angiogenesis and MDA scores were significantly higher in G2 than in G3.

Conclusion: Daily oral vitamin C was found to be effective in preventing intraabdominal adhesion formation in the rat uterin horn model.
