The Effects of PET/CT Application on TRPM2 and Apoptosis in Rat Brain Tissue

Objective: In this study it was aimed to examine the immunoreactivity of Transient Receptor Potential Melastatin 2 (TRPM2) ion channels which has a known interaction with oxidative damage in the brain tissues of rats undergoing PET/CT.

Material and Method: In this study 20 adult Sprague Dawley female rats were used. The rats were divided in to 4 groups. Group I was used as control. On 1st day of experimental study computed tomography administered to the second group, to the third group a single dose 1mci FDG was injected and PET/CT was performed after 1 hour. To the fourth group the single dose of 1mci FDG was injected on 1st day of the experimental study. Rats were decapitated and brain tissues were removed. Routine follow up was performed and tissues were embedded in paraffin blocks. Sections taken from the blocks were administrated tunnel staining for apoptozis and avidin-biotin-peroxidase method for TRPM2 and MDA. Tissues were examined in the light microscope and photographed and then statistical analysis was performed.

Results: When compared with the control group, in all other groups no significant differences were determined in brain tissues for apoptotic index, TRPM2 and MDA immunreactivities.

Conclusion: In this study, we concluded that, PET/CT imagining doesn’t cause apoptosis in rat brain tissue, moreover there is no significant effect on TRPM2 ion channels and MDA expressions.
