Investigation of the Potential of the Functional Conjugate System in the Diagnosis of Gaucher Disease

Objective: It is aimed to synthesize hybrid conjugate structures functionalized with specific antibodies under different conditions by using organic-inorganic hybrid synthesis method, to elucidate the resulting structures, to characterize their properties, and to monitor the binding of these structures to target molecules specific to Gaucher disease, which is difficult to diagnose with the ELISA test system.

Material and Method: β-glucocerebrosidase specific polyclonal antibody (Anti-GBA 3) was chosen as the binding antibody, β-Glucosidase enzyme was used as standard with the Sandwich ELISA method in the concentration range of 12.5-400 ng/mL. The test was made visible by adding antibody-enzyme-inorganic hybrid conjugate. Thus, the level of ß-Glucosidase in the sample could be captured with the combination created.

Results: While Anti-GBA 1 conjugate gave a high response in the conventional sandwich ELISA method, Ab 1 (10 μl)-HRP-inorganic hybrid conju-gate gave a higher response in the hybrid system. In the classical method, the linear analytical performance is determined as R2 >0.99 and the LOQ value is set at 10 ng/mL, whereas in the hybrid conjugate system, R2 >0.99 and the LOQ value is determined as 12.5 ng/mL.

Conclusion: Compared to the traditional ELISA method, the hybrid system allows for the one-step binding of the HRP enzyme to the conjugate, resulting in a conjugate structure with enhanced capture capability and catalytic activity.
